Karthi also worked as a graphic designer in New York for short period part-time.

He started his career as Assistant Director under Mani Ratnam and then he offers the lead role in his debut movie Paruthiveeran in 2007, which gave him more critics, and also he won Filmfare Award for Best Male Debut. Then he appeared in his next blockbuster movie Aayirathil Oruvan (2010), based on an action-adventure film directed by Selvaraghavan.

Then he gave one-by-one commercial hit movies and these are, Paiyaa (2010), Naan Mahaan Alla (2010), and Siruthai (2011). This movie gave not a good response to the box office but after that, he gave numerous blockbusters, Madras (2014), Thozha (2016), Theeran Adhigaaram Ondru (2017), Kadaikutty Singam (2018), and Kaithi (2019).
